Strong expression is seen predominantly in the muscles of the pharynx, the pharyngeo-intestinal valve, and in cells surrounding the rectum, which appear to be the anal depressor muscle and the intestino-rectal valve. Other expression is occasionally seen in the vulval muscle cells (vm1 and vm2), bodywall muscle cell nuclei, and cells within the tail spike. Expression is seen from late embryo to adulthood. |

The second component is in the vm2 vulval muscles. The expression here is cytoplasmically localised, and covers only the period around the L4 to adult molt This gene has two distinct modes of expression. The earlier component is in a subset of cells of the pharynx, and is nuclear localised. Expression here is from late embryogenesis onwards, the subset consisting of the m4 muscles in the mid metacorpus,the m2 muscles and e1 and e2 epithelial cells of the procorpus, and the m8 muscle of the posterior terminal bulb. |

The mls-1 transgenes were expressed in the sex myoblast lineage and in a subset of the enteric muscles. Expression of the mls-1 transgenes in the M lineage was detected as early as the sex myoblast granddaughters (M.vl/rpaaa/pa/p). The transgene product was initially observed to accumulate in the uterine muscle progenitors and persisted throughout uterine muscle differentiation. In the initial period after segregation of uterine muscle and vulval muscle fate, the vulval muscle progenitor lineage lacked mls-1 transgene activity. However, after division of the vulval muscle progenitors to Type 1 and Type 2 vulval muscle precursors, mls-1 transgenes were expressed in Type 2 vulval muscles. Expression of mls-1 transgenes was not observed in Type 1 vulval muscles. mls-1 transgenes were also expressed in a subset of the enteric muscles. mls-1 was expressed in the left intestinal muscle (AB.plpppppaa), right intestinal muscle (MS.ppaapp), and the anal depressor muscle (AB.plpppppap). Expression was not detected in the sphincter muscle (AB.prppppap). |

In the pAB::GFP fusion, expression was seen in some pioneering neurones of the nerve ring, beginning at the early comma stage. At the two-fold stage, expression was detected in some 10 neurones in the head that extend axons into the nerve ring, and in two neurones in the tail that extend processes anteriorly. This expression pattern was confirmed by immunohistochemistry with MAb 16-48-2. At the three-fold stage, expression was seen in all DA motoneurones and persisted while they pioneered the dorsal nerve chord. It was also seen in four to six neurones in each of the four head ganglia, including ALA and RID in the dorsal ganglion, and four of the six neurones of the terminal bulb, including M5. In the tail, two neurones in the pre-anal ganglion and six in the lumbar ganglion, including PVQL and PVQR, showed pAB::GFP expression. Additionally, a transient expression was seen in the four rows of bodywall muscle cells in the embryo. After hatching, in L1 larvae, the expression domain extended to amphid and phasmid socket cells, and subsequently in L2 larvae to all the newly born AS motoneurones. In hermaphrodite L3 larvae, expression was seen in the sex myoblasts subsequent to their anterior migration towards the position of the presumptive vulva, and in adult worms at a high level in the vulval muscles vm1 and vm2. In males, expression was seen in the diagonal and spicule retractor muscles. |
