Subcellular Localization  gamma-Tubulin was detected at the centrosomes of mitotic germ cells localized at the distal tip of the gonad. In meiotic cells arrested at the pachytene stage in the middle of the gonad, g-tubulin staining of the centrosomes was more difficult to visualize. In oocytes arrested in diakinesis of prophase I in the proximal gonad, only a diffuse cytoplasmic g-tubulin staining was detected and no centrosomes. Immunostaining of mature oocytes failed to reveal any g-tubulin at the poles of both meiosis I and meiosis II spindles. The spermatheca of the hermaphrodite gonad and identified spermatocytes undergoing meiotic division was examed. g-Tubulin staining was detected at the poles of the spindles as focused dots corresponding to the centrosomes. Mature sperm cells were also observed, showing a weak dot staining at the position expected for the centrioles.
