Primary Identifier  Expr4351 Remark  The distribution of the chromosome paints and DPY-27 antibody can be seen more clearly in the large gut nuclei of the adult hermaphrodite. These nuclei are 32-ploid, yet both the DPY-27 antibody staining and chromosome paints are nonetheless seen in restricted domains. By contrast, in favorable optical sections in which the chromosome paint for chromosome Ill and DPY-27 can both be seen, the two do not colocalize. These results were confirmed in a separate experiment with gut nuclei, in which the nuclear location of DPY-27 was detected by indirect immunofluorescence with fluorescein-labeled antibodies, the X chromosome was detected by hybridization of rhodamine-dUTP-labeled paint, and chromosome Ill was detected by hybridization of a Cy5-dCTP-labeled paint. Comparison of the combined DPY-27 and X chromosome images, the combined DPY-27 and chromosome Ill images, and the combined DPY-27, X chromosome, and chromosome Ill images reveals that the X paint and DPY-27 colocalize but occupy a different domain from that of chromosome Ill.
Subcellular Localization  DPY-27 antibody staining and X chromosome paint are colocalized. In contrast, DPY-27 antibody staining (green) and the chromosome paint for the middle of chromosome III (red) were not colocalized in embryos.
