    Expr11526 CYP33E2 promoter-driven expression of GFP occurred exclusively in the pharynx and, not visible in each individual, in the pharyngeal-intestinal valve of the nematodes. This type of strong pharynx-restricted expression was observed throughout larval development and in adult nematodes. Expression was most prominent in the pharyngeal pro- and meta-corpus. Confocal imaging suggested marginal, muscle, and/or epithelial cells as the major expression sites of the pCYP33E2::GFP construct within the pharynx. Radially, only marginal cell types are continuously organized with three-fold symmetry around the pharyngeal lumen. Imaginary cross sections derived from confocal imaging series of the pro- and meta-corpus indicated that the GFP reporter was expressed in the three marginal mc1 cells, but not in the pm3 and pm4 muscle cells. A further labelling of the mc2 and mc3 marginal cells in the isthmus and terminal bulb becomes then visible. Expression was observed in finger-like fluorescent structures that represent the interlocking extensions that hold marginal cells to muscles. Furthermore, an expression of pCYP33E2::GFP also in the epithelial e1, e2, and e3 cells seems most likely. Fluorescence might also correspond to pm2 muscle cells.

Picture: Fig 2A to 2G.   Expr9051 In wild-type embryos, ten-1a::gfp is first expressed in a cluster of cells in the anterior half at approximately 150 minutes after fertilization. These cells are precursors to the hypodermal cells, which are evident at 300 minutes post-fertilization, when the cells intercalate and begin the process of ventral closure, and to pharyngeal and intestinal cells, which are evident beginning at the bean stage. In later stages, strong expression of ten-1a::gfp persists in pharyngeal and intestinal cells, and appears in several head neurons. Examination of L1 larvae and adults allowed us to identify 8 pharyngeal cells that express ten-1a::gfp: the three marginal cells mc1, the three marginal cells mc3, and the neurons M2L and M2R. Adults also express ten-1a::gfp in vulva muscles, the gonad distal tip cells, the intestine, several tail neurons including DVB and some other cells.  
    Expr11324 Fairly weak expression, pharynx: pm3,4,6,7,8 mc1,3, glands (strong), excretory cell, hypodermis (borderline detectable).  
Picture: Fig 6, Fig 7.   Expr8860 SMF-2::GFP was mostly restricted to the mc1, mc2, mc3 epithelial cells of the pharynx and the pharyngeal-intestinal valve cells vpi1 to 6, displaying an anterior-posterior expression gradient. SMF-2 was also observed in the gonad sheath cells at the adult stage. The GFP signal was detected from late embryogenesis to adult stage, with generally higher expression levels in young larvae (L1 stage). SMF-2::GFP was associated with intracellular cytoplasmic compartments and to a lesser extent to the apical membrane of the mc epithelial cells.
    Expr2624 Promoter-GFP transgenes indicated that lam-3 is expressed during gastrulation and through embryogenesis in pharyngeal, intestinal and epidermal cells. In the larvae, lam-3 gene expression is maintained in the spermatheca and in the pharyngeal m3-m8 and mc cells.
