15 Expression Patterns

Remark Reporter Gene Primary Identifier Pattern Subcellular Localization
Expr11526 CYP33E2 promoter-driven expression of GFP occurred exclusively in the pharynx and, not visible in each individual, in the pharyngeal-intestinal valve of the nematodes. This type of strong pharynx-restricted expression was observed throughout larval development and in adult nematodes. Expression was most prominent in the pharyngeal pro- and meta-corpus. Confocal imaging suggested marginal, muscle, and/or epithelial cells as the major expression sites of the pCYP33E2::GFP construct within the pharynx. Radially, only marginal cell types are continuously organized with three-fold symmetry around the pharyngeal lumen. Imaginary cross sections derived from confocal imaging series of the pro- and meta-corpus indicated that the GFP reporter was expressed in the three marginal mc1 cells, but not in the pm3 and pm4 muscle cells. A further labelling of the mc2 and mc3 marginal cells in the isthmus and terminal bulb becomes then visible. Expression was observed in finger-like fluorescent structures that represent the interlocking extensions that hold marginal cells to muscles. Furthermore, an expression of pCYP33E2::GFP also in the epithelial e1, e2, and e3 cells seems most likely. Fluorescence might also correspond to pm2 muscle cells.
Expr11290 Eeak expression, pharynx: pm6-8, mc1-3, some glands, pharyngo-intestinal valve, gut, rectal glands, body wall muscle (adult only), in young larvae: unidentified head cell (head mesodermal cell?), CAN.
Expr14587 The integrated smoc-1 transcriptional reporter showed strong GFP expression. GFP was first detectable in several cells located in the anterior of bean stage embryos. In the developing larvae, GFP is expressed in cells of the pharynx, the intestine and the posterior hypodermis. Pharyngeal cells expressing smoc-1p::gfp include the epithelial cells e2, the marginal cells mc1 and mc2, the M4 neuron, and all six of the pharyngeal/intestinal valve cells. Cells of the posterior hypodermis expressing smoc-1p::gfp include hyp8, hyp9, hyp10, and hyp11. Expression in these tissues persisted from the L1 larval stage through adulthood. We noted that while all transgenic animals showed GFP expression in the pharynx and the posterior hypodermis, a small fraction of animals (~8%) did not exhibit GFP expression in all or some of the intestinal cells. We observed no GFP expression in any other tissues, including the nerve cord, body wall muscles (BWMs), or the M lineage.
Expr16520 Ppa-nas-6 was expressed in the pharyngeal muscles (pm2), arcade cells, epithelial cells (e1, e2, e3), marginal cells, (mc1, mc2, mc3), amphid neurones, and the pharyngeal-intestinal valve.

Weak or rare expression in pm4, pm5. Expression in the nervous system: DDn, DVA, DVB, DVC, PVP. Expression in the reproductive system: In adult stage, expressed in proximal gonad sheath, spermatheca. In developing larva stage, expressed in uterus, spermatheca. inx-10 is localized to pharyngeal precursors from early stages of embryogenesis, and by three-fold stage, all pharyngeal muscles except pm4 are seen to express it at high levels.
Expr11259 pax-1::HIS-GFP is expressed in the nuclei of all three groups of marginal cells, pm8 and v1.
Picture: Fig 6, Fig 7.   Expr8860 SMF-2::GFP was mostly restricted to the mc1, mc2, mc3 epithelial cells of the pharynx and the pharyngeal-intestinal valve cells vpi1 to 6, displaying an anterior-posterior expression gradient. SMF-2 was also observed in the gonad sheath cells at the adult stage. The GFP signal was detected from late embryogenesis to adult stage, with generally higher expression levels in young larvae (L1 stage). SMF-2::GFP was associated with intracellular cytoplasmic compartments and to a lesser extent to the apical membrane of the mc epithelial cells.
Expr2624 Promoter-GFP transgenes indicated that lam-3 is expressed during gastrulation and through embryogenesis in pharyngeal, intestinal and epidermal cells. In the larvae, lam-3 gene expression is maintained in the spermatheca and in the pharyngeal m3-m8 and mc cells.

Expr13074 All the invertebrate lysozymes have in common an intestinal expression pattern. However, one subset appeared to have additional pharyngeal expression whereas the other exhibited some neuronal promoter activity. ilys-1 expressed in the pm3 cells in the procorpus, the pm4 cells in the anterior bulb pharyngeal muscles (sieve) and the marginal cells mc1 and mc2.
Other strain-- UL820   Expr154 Expression is seen in the marginal cells of the pharynx, from early larval stages until the adult stage. In early larvae the mc1 and mc2 marginal cells show expression, whereas in adults only the mc2 marginal cells are seen to express.

Expression in the nervous system: AVD, AVK, RIS, URB. Pharyngeal and neuronal expression of inx-6 start around threefold stage, and some of the expression in head and tail neurons disappears after L1 stage.
Expr16521 Cel-nas-6 is expressed in pharyngeal muscles, mc2 marginal cells, and hypodermal tissues (Park et al. 2010). We also generated C. elegans transgenic lines expressing Ppa-nas-6::T2A::Venus under the control of the Cel-nas-6 promoter to confirm the expression pattern of Cel-nas-6. Our reporter also showed the expression in hypodermis and pharyngeal muscles (pm2, pm6) and weak expression in mc2 marginal cells. In C. elegans, the nas-6 expression is not detected in arcade cells, epithelial cells, mc1/mc3 marginal cells, head neurons, and the pharyngeal-intestinal valve.
Picture: N.A.   Expr8909 Expressed in pharyngeal muscle, mc2, all intestinal cells, major hypodermis and body wall muscle.
Expr13079 The short ilys-3 promoter drove GFP expression mainly in the pharynx grinder muscles pm7, the isthmus marginal cell mc2 and muscle cell pm5. In addition, all intestinal cells exhibited ilys-3 promoter activity, with some variability. Intestinal expression of ilys-3 appeared to be temporally regulated. GFP was first detected in intestine of L1 hatchlings but declined at the L2 transition. By the L4 stage, the intestinal GFP signal became stronger and continued through adulthood. The intensity of GFP was very high in aging adult worms that were no longer self-fertile. Whereas the intestinal expression of ilys-3 changed during larval growth and adult life, the pharyngeal GFP reporter expression remained constant in larval and adult stages. Weak epidermal promoter activity was also detected. The long ilys-3 promoter revealed additional GFP expression in the six scavenger coelomocyte cells, but at a significantly lower level in other tissues.
