    Expr1777 CPL-1 was localized in various cells in early to late embryonic stages as well as in the eggshell surrounding the embryos. CPL-1 accumulated in numerous cells during early embryogenesis; however, as embryos progressed through middle and late developmental stages, the CPL-1 protein clustered in gut cells of the embryos at the early L1 stage. In addition, Ce-CPL-1 was present in the hypodermal and cuticular regions in all larval and adult stages, in the pharyngeal lining of all stages, and in the pharyngeal gland of the adult worms only. The subcellular localization of the native CPL protein in C. elegans was determined by immunoelectron microscopy staining. In larval and adult stages the native protein was localized in the cuticular region of the worms and on the outer cuticle of the ridge of the annuli. In molting larvae, the protein was localized in both the old and new cuticle. In addition, the protein was localized on the surface of late-stage embryos.
    Expr2933 SRP-2 expression is evident in numerous cells of the C. elegans embryo in the early stages of development (36-100-cell stage). However, in the later stages of embryonic development, expression is confined to a few cells of the anterior hypoderm and the eggshell. In the L1 and L2 larvae, expression is seen in a number of hypodermal (possibly hyp 1, 2, and 3) cells near the buccal cavity. Expression is also seen in the sensory support (sheath or socket) cells of several sensilla. Strong expression is visible in the posterior intestinal cells, seam cells, and the body hypoderm. In the adult hermaphrodite, strong punctate expression is seen in the hypodermal cells surrounding the anterior and posterior bulb of the pharynx. Expression was also seen in hyp 7 cells near the vulval opening. A strong striated staining pattern was seen in what appears to be the fibrous organelles that link muscle/hypoderm/cuticle. GFP expression was also observed in the phasmid (PHA and PHB) neurons, which was confirmed by co-staining with the amphid neuron-specific dye, DiI. The overall SRP-2::GFP expression pattern was similar in the hermaphrodites and males.  
    Expr13979 DDO-3::mCherry was localized in the excretory pore cell, head tip cells, and hypodermal cells. On the other hand, DDO-3::mCherry was not localized in proximal gonadal sheath cells at any point during development, suggesting that DDO-3 is secreted from these cells in a SP-dependent manner. Interestingly, in the adult stage, DDO-3::mCherry was localized on the surface of the oocyte, which is surrounded by gonadal sheath cells, suggesting that DDO-3 produced in and secreted from proximal gonadal sheath cells is transferred to the oocyte surface. Furthermore, DDO-3::mCherry was also localized in the eggshell within the adult uterus, as well as in the uterine space, and it remained localized in the eggshell until hatching. In hermaphrodites, in addition to the excretory pore cell, head tip cells, hypodermal cells, oocyte surface, uterine space, and eggshell, DDO-3::mCherry was also present in coelomocytes from the late embryonic stage, and accumulated in vesicle-like structures at least from the adult stage.

Expression of the CPL-1::GFP fusion protein was also observed along the length of the pharyngeal lining in L1-L4 stages, but was restricted to the posterior bulb of the pharynx in adult worms. Furthermore, robust expression was observed in the eggshells surrounding the embryos inside the hermaphrodite and in the regions of the vulva, uterus, and spermatheca. In contrast to the transcriptional reporter construct there was no CPL-1::GFP expression in the gut. GFP was detected in the eggshells of laid eggs in different stages of development.
    Expr13069   CPG-2 localizes to the inner layer of the trilaminar eggshell. CPG-2 localizes to both the CPG layer and to the perivitelline space.
    Expr13388 mNG::3xFLAG::CPG-1 is expressed in the cytoplasm of oocytes and in the eggshell of embryos.
