• Pattern: The anti-LIN-56 antibodies were used for whole-mount staining of worms. Starting at the late one-cell or early two-cell stage, LIN-56 staining was observed in the nuclei of most if not all somatic cells throughout embryonic and larval development as well as adulthood. LIN-56 is present in the P(3-8).p vulval equivalence group and their descendants throughout vulval development. Germline staining was observed in both wild-type and lin-56(n2728) animals and thus was not specific.

• Subcellular Localization: To confirm that LIN-56 is localized to nuclei, subcellular fractionation of embryo lysates was performed. Lamin was used as a marker for nuclear fractions and SQV-4 as a marker for cytosolic material. The majority of the LIN-56 protein co- fractionated with nuclear lamin; LIN-56 protein was barely detectable in the SQV-4-containing cytosolic fraction. LIN-56 is thus stably associated with nuclei.
