GFP::ALG-5 appeared cytoplasmically diffuse but also formed distinct puncta at the nuclear periphery reminiscent of P granules, a germ cell-speci c class of RNA granules that function in mRNA surveillance. P granules contain the piRNA-associated Piwi protein, PRG-1, and much of the siRNA pathway machinery. GFP::ALG-5 foci overlapped with the P granule marker RFP::PGL-1 foci, indicating that, similar to many known piRNA and siRNA components, ALG-5 localizes to P granules.
