| Subcellular Localization | Using affinity-purified antibodies, FZY-1 was detected in early-stage embryonic cells at prometaphase and metaphase of mitosis and meiosis I. At prometaphase, highly condensed chromosomes were seen, and FZY-1 was localized around condensed chromosomes. At metaphase, FZY-1 localized along chromosomes aligned at the metaphase plate. After the separation of sister chromatids, the FZY-1 signal disappeared from the chromosomes, and at anaphase and interphase, cytoplasmic staining was observed. FZY-1 was observed around the meiotic chromosomes in cells at metaphase of meiosis I, but no signal was detected in cells at anaphase of meiosis I or meiosis II. The FZY-1 signal in cells at prometaphase or metaphase was not detected when antibodies were preincubated with purified recombinant FZY-1 protein, a finding that confirms the specificity of the antibodies. |
