| Subcellular Localization | GFP-fusion constructs of CeCaM-KI was transfected into COS-7 cells. Expression of the GFP-fusion protein of each CaM-KI was confirmed by Western blotting using anti-GFP antibody and the CaM overlay method. CeCaM-KI (wild type, Pro2-Ala348) is highly concentrated in the nucleus. When a mutant CeCaM-KI lacking 6 residues (Pro2-Arg7) at the NH2-terminal region was used, it was no longer staying in the nucleus, suggesting that the NH2-terminal 6 residues contain a potential NLS. GFP CeCaM-KK was detected localized in both the cytoplasm and the nuclei of transfected cells. |
