One or two large areas of signal were observed at the center of cells, the nature of which is unknown. A punctate distribution was also observed in elongating filopodia-like structures. There was no indication of preferential protein accumulation at the ventral or dorsal sides; rather, the fluorescent signals were uniformly distributed at the cell margin. Most GFP foci were observed in regions distinct from RAB-5-positive early or late endosomes, as labeled with mCherry::RAB-5. Thus, RIN-1 protein probably accumulates on, or just beneath, the plasma membrane during these stages of development. |
