Some colocalization with endogenous EEA1 was seen in non-nonrestored images, but it was mostly in the perinuclear region where the cytoplasmic volume is largest and was not apparent in restored images where structures that reside in close proximity can be resolved from each other . Analysis of the 3D images revealed a more peripheral localizationof endosomes containing WDFY2 compared to those enriched in EEA1, suggesting that vesicles containingWDFY2 are closer to the plasma membrane that those containing EEA1. The pattern suggests the possibility that WDFY2 may mark a subset of endosomes that serves as a first stage of endocytic transport, operating between the plasma membrane and EEA1-enriched endosomes.
