While DMA-1::GFP is dominantly expressed in PVD and FLP, especially by the adult stage, DMA-1::GFP expression in additional head and ventral cord neurons was detected during larval and early adult stages. DMA-1 expression was detected in the PVD cell body shortly after PVD are born (L2 larval stage) before any dendrite outgrowth or branching has occurred. Weaker DMA-1::GFP expression can be seen in ventral cord motorneurons and in the sub-lateral cords.Expression was variable from animal to animal, seen in both commissural and non-commissural motorneurons, and was mostly undetectable by the early adult stage. In the head, expression of DMA-1::GFP was detected during development in additional head neurons besides FLP. Of these neurons, the strongest expression was detected in neurons that run around the nerve ring and extend processes along the sub-lateral cords (likely some combinationof the SIA, SIB, SMB and/or SMD classes of neurons). A number of additional head neurons displayed inconsistent DMA-1::GFP expression from animal to animal. Again, expression in these additional neurons was transient and mostly undetectable by the early adult stage. d, Strong expression DMA-1::GFP was detected in vulva cells during the L3-L4 larval stages. |
Bright DMA-1::GFP localization can be seen in intracellular membrane structures that are likely to be golgi/ER. The plasma membrane is also clearly visible, indicating that DMA-1::GFP is a cell-surface protein. |

Expression observed in most or all neurons at high levels as well as in some hypodermal and muscle cells. In most neurons, sax-3::GFP expression is transient, peaking during the period of axon outgrowth. Highest level of expression is observed in the embryo, particularly during the initial period of axon outgrowth at 350-400 min (comma-stage). At comma-stage high level of expression is observed in anterior embryo, including most developing neurons of the nerve ring and a swath of ventral cells that includes the developing motor neurons of the ventral nerve cord and posterior neurons such as PVQ. Lower level of expression is observed in the epidermal cells. 200-400 - cell stage; 200-300 min of development: Expression seen in all epidermal cells at low level. 3-fold stage; >500 min of development: Expression seen in muscles that extend from nerve ring to the anterior tip of the head. L1: Expression in motor neurons in the head including RMD, RMG, SMD, SIA and SIB neurons; projection interneurons in the head and tail, including AVA, AVB, PVC, AVD, PVQ and ALA neurons; and the sensory OLQ neuron. This neuronal expression diminishes throughout postembryonic development. During L1, expression persists in the head muscles and appears in muscles along the body wall, with ventral muscles expressing more strongly than posterior muscles. This expression continues until the adult stage. Epidermal expression was rarely seen in larval stages. L2: Expression observed in HSN motor neurons. HSN motor neurons extends a growth cone (axon from their lateral cell bodies) towards the ventral midline and expression is at its highest at this point. L3: Expression observed in the axon reaching the ventral nerve cord. L4: Expression continues in HSN, when HSN axon grows anteriorly to the head. Adult: Expression decreases in HSN at this stage after the completion of HSN axon outgrowth. In addition expression observed in motor neurons, interneurons and sensory neurons listed above also postembryonic ventral cord motorneurons, some interneurons from the tail and head, body wall and vulval muscles. |

UNC-42 expression was detected with a mouse antiserum in several cells as early as 260 minutes of embryogenesis and in neurons at high levels in the head by comma stage, the time when these neurons extend axons and establish connections. Expression in head neurons continues into adulthood. UNC-42 was strongly expressed in at least 20 pairs of neurons of the head, including the AVA, AVD and AVE interneurons, ASH sensory neurons, and RMD and SMB motor neurons. Other neurons that express high levels of UNC-42 include the AIN, AVH, AVJ, AVK, RIV, SAA and SIB interneurons. The same expression pattern in the head was detected in animals that carry a transgene that encodes an UNC-42 protein tagged at its C terminus with GFP(gmEx186). Antiserum to UNC-42 and the gmEx104 GFP reporter gene revealed low levels of unc-42 expression in hypodermis and additional neurons in the head. Transient expression of UNC-42 protein was also detected in the DD motor neurons at hatching and at low levels in postembryonic ventral cord motor neurons derived from P11. UNC-42 protein was not detected in descendants of P2-P10 or P12. In the tail, unc-42-gfp (gmEx104) is strongly expressed in PVT. |
