Primary Identifier  Expr9296 Subcellular Localization  In worms expressing a GFP::STA-2 fusion reporter in the epidermis (pcol-12::GFP::STA-2), a fluorescent signal was clearly seen in the nucleus but also in bands underneath the cuticle and in some vesicles. Equivalent patterns were also obtained when STA-2 was tagged with DsRed2 or mCherry and when the fusion protein was expressed under the control of the sta-2 promoter (psta- 2::GFP::STA-2). As the different chimeric proteins were functional, the localization seen with these reporter proteins is likely to reflect the distribution of the endogenous protein. Using the mCherry::STA-2 construct,we found that the fluorescent signal colocalized with a clathrin heavy-chain CHC-1::GFP reporter protein, a specific marker of newly formed endocytic vesicles (Greener et al., 2001; Sato et al., 2009).
