• Pattern: We found that in the adult nervous system unc-42 is strongly and consistently expressed in 40 neurons located in the head of the worm that fall into 15 anatomically distinct neuron classes. At the first larval stage, expression is detected in the same set of neuron classes as observed in the adult, with the addition of very weak and inconsistent expression in AVJ and SIA that disappears by the adult stage. In the embryo, expression is first observed in a few neuroblasts before the bean stage. At the bean stage, when most neurons have terminally divided, unc-42 expression commences and reaches the full complement of unc-42(+) cells at the 1.5-fold stage. The cellular sites of expression in the embryonic nervous system appear to be the same as observed post-embryonically, a notion further supported by recent scRNA data (Packer et al., 2019). No expression is observed outside the nervous system.

• Pattern: Using a fosmid-based GFP translational fusion, we found that zag-1 was expressed in the six TRNs but not in the FLP and PVD neurons. In addition to TRNs, zag-1 was expressed in the AIB, AIM, AIN, AIZ, AVA, AVB, AVD, AVE, AVG, AVK, AVL, M4, M5, RIA, RIB, RIF, RIG, RIM, RIV, RMD, RME, RMF, RMH, SIA, and SMD neurons in the head, all the DD, VD, and VC neurons in the ventral cord, and the DVA, DVB, LUA, PDA, PVC, PVP, PVQ, PVR, and PVT neurons in the tail. zag-1 is also expressed in the serotonergic HSN neurons.

As in vertebrates, sem-2 expression was observed in postmitotic neurons, but in contrast to the broad expression of SoxC genes in vertebrate postmitotic neurons, only a single class of postembryonic neurons expresses sem-2, the RMH class. sem-2 expression in these neurons is observed throughout larval and adult stages. We did not detect any sem-2 expression in the RME neurons themselves, but sem-2 is instead expressed in the progenitor of RMEL/R.

Unfortunately, attempts to colocalize the full length proteins were unsuccessful using this approach as SER-1::YFP and SER-1::CFP aggregated significantly when expressed in vulval muscle.
