Remark Reporter Gene Primary Identifier Pattern Subcellular Localization
  [lin-42ap::gfp-pest] transcriptional fusion. The start codon of lin-42a and 3,043 bps of upstream sequence were fused with gfp-pest. --precise ends. Expr9861 GFP expressed from the lin-42a promoter was robustly expressed in the pharyngeal myoepithelium midway through the L1 stage and continuously thereafter. The lin-42a reporter was also expressed in hyp7 and the lateral seam cells of late L1-stage larvae undergoing the first molt. Expression of the lin-42a reporter in the hypodermis was detected later in the L1 stage than expression of the lin-42b reporter. Fluorescence from the lin-42a reporter also persisted through ecdysis, whereas fluorescence from the lin-42b reporter dissipated during lethargus. To fully define the cyclical expression pattern of the lin-42ap::gfp-pest fusion gene, we measured the relative intensity of the associated fluorescence in the hypodermis throughout larval development. The intensity of fluorescence repeatedly increased 2- to 10-fold during the molts and dropped to background levels early in the next life stage.  
    Expr12246 lin-4 was temporally expressed in the hypodermal seam cells at the early L2 stage and expression persisted in adulthood. Furthermore, the spatial patterning of lin-4 in the hypodermal seam cells correlated with the requirement of this gene at the L1/L2 transition to direct normal seam cell development.lin-4 expression was first observed within the developing gonad at the beginning of L3, specifically in the distal tip cells and the anchor cell of the somatic gonad and expression in the distal tip cells continued in the adult. lin-4 and let-7 were weakly expressed in the anchor cell, which is required for proper vulval patterning, and in the P5.p, P6.p, and P7.p cells that will later differentiate into the mature vulva.  
    Expr11055 An nhr-165::gfp translational reporter shows nuclear expression in the lateral hypodermis (hyp7, but not hyp8-11) near the time of larval molts. This expression is highest and seen in the largest number of cells in the tail prior to the L3-L4 molt.  
    Expr13927 In addition to recapitulating previously reported expression patterns [in the gonad from mid-L1 larval stage until around the L1/L2 molt in both sexes and from the L3 larval stage through adult in the hermaphrodite gonad in the spermatheca and sheath cells], this new construct revealed expression of FKH-6::GFP in 2-4 male gonadal cells in the L3 larval stage. This is the first report of FKH-6 expression in males after the L1/L2 molt. The expression of FKH-6::GFP in the gonad of the L3 male is reproducible, appearing in 54 of the 58 male animals observed during this time period, but the expression is short-lived, and coincides with the period when the proximal region of the migrating male gonad is extending past the distal tip of the gonad.
