Immuno-EM on embryos from wild-type hermaphrodites prepared by high-pressure freezing/freeze substitution confirmed the result. In sections containing centrioles, gold particles were observed associated with the centriolar cylinders. Immuno-EM for SAS-5 indicates that it too localizes to the centriolar cylinders. The combination of light microscopy and EM analysis indicates that SAS-6, like SAS-4 and SAS-5, is a bona fide centriolar protein.

Cold-induced depolymerization of the microtubule network does not alter SAS-4 distribution at the centrosome. Transgenic animals expressing GFP-SAS-4 were generated. Staining of transgenic embryos with anti-GFP antibodies confirmed the findings made with anti-SAS-4 antibodies.   Expr2384   Anti-SAS-4 antibodies label a tiny dot in the center of centrosomes, as well as in the cytoplasm, to a lesser extent. Both centrosomal and cytoplasmic staining are reduced by ~60% in sas-4(RNAi) embryos, indicating that RNAi only partially depletes SAS-4 but that both localizations reflect bona fide SAS-4 distribution. SAS-4 is present at centrosomes throughout the cell cycle in early embryos. While a single dot can be detected per MTOC during most of the cell cycle, two dots become visible per MTOC toward the end of mitosis. As this coincides with the onset of centrosome duplication, the appearance of two dots likely reflects splitting of the mother/daughter centrioles. Furthermore, a tiny dot was observed next to the male pronucleus immediately after fertilization, suggesting that SAS-4 is present with the paternally contributed centrioles. Staining of sperm confirmed that SAS-4 is present as a tiny dot in the perinuclear region, where centrioles are known to reside. In fact, a tiny dot of SAS-4 is present in male germ cells throughout spermatogenesis.
