WormBase Gene ID  ? WBGene00006592 Gene Name  dpy-31
Sequence Name  ? R151.5 Brief Description  dpy-31 encodes an astacin zinc-metalloprotease of the BMP-1 (bone morphogenetic protein-1)/TOLLOID family; dpy-31 activity is required for proper cuticle formation and thus for normal body morphology; in addition, dpy-31 is essential for embryonic and larval development at 25 degrees C; genetic and immunofluorescence studies suggest that DPY-31 functions as an endopeptidase that is required for processing, and ultimately proper extracellular assembly, of the SQT-3 cuticle collagen; dpy-31 is expressed in most hypodermal cells, with the exception of larval stage seam cells, throughout the life cycle with particularly strong expression seen during embryonic and larval stages; dpy-31 is also expressed in some head neurons, the amphid socket cells, rectal epithelial cells, and vulval epithelial cells in L4 larvae and adults.
Organism  Caenorhabditis elegans Automated Description  Predicted to enable metalloendopeptidase activity. Involved in cuticle development involved in collagen and cuticulin-based cuticle molting cycle. Predicted to be located in extracellular region. Expressed in several structures, including amphid sheath cell; excretory duct; hypodermis; socket cell; and vulva.
Biotype  SO:0001217 Genetic Position  III :-0.771±
Length (nt)  ? 5459

Strain: BC12765 [toh-2::gfp] transcriptional fusion. PCR products were amplified using primer A: 5' [AAGGTTATAAGCTCCGCCTATTG] 3' and primer B 5' [TTTTGTGGATGCTGAAATTAAAGT] 3'. Expr6546
dpy-31 in this paper = toh-2 (WS145)   Expr3138 A version of the pD31P3.4G construct devoid of NLS (pD31P3.4G-N) confirmed this expression pattern. In transgenic animals carrying pD31P3.4G-N, GFP was detected in most hypodermal cells, except in the seam cells of larval stages. Expression of DPY-31 is particularly pronounced at all life stages in the amphid socket cells at the anterior end of the animal and in the rectal epithelial cells at the posterior end. In L4 and adults, DPY-31 expression was also seen in the vulval epithelial cells. Fluorescence was also observed in some head neurons, although the identity of such neurons has not been determined. Transgenic worms carrying pD31P3.4G express GFP in most hypodermal nuclei throughout the life cycle. Expression was first detected in embryos at the twofold stage. The GFP signal was particularly intense in embryos and early larvae.

    Expr10273 Inferred expression. EPIC dataset. http://epic.gs.washington.edu/ Large-scale cellular resolution compendium of gene expression dynamics throughout development. This reporter was inferred to be expressing in this cell or one of its embryonic progenitor cells as described below. To generate a compact description of which cells express a particular reporter irrespective of time, the authors defined a metric "peak expression" for each of the 671 terminal ("leaf") cells born during embryogenesis. For each of these cells, the peak expression is the maximal reporter intensity observed in that cell or any of its ancestors; this has the effect of transposing earlier expression forward in time to the terminal set of cells. This metric allows straightforward comparisons of genes' cellular and lineal expression overlap, even when the expression occurs with different timing and despite differences in the precise time point that curation ended in different movies, at the cost of ignoring the temporal dynamics of expression, a topic that requires separate treatment. For simplicity, the authors use the term "expressing cells" to mean the number of leaf cells (of 671) with peak expression greater than background (2000 intensity units) and at least 10% of the maximum expression in that embryo. Quantitative expression data for all cells are located here: ftp://caltech.wormbase.org/pub/wormbase/datasets-published/murray2012/
    Expr2011072 Single cell embryonic expression. Only cell types with an expression fraction of greater 0.2 of the maximum expressed fraction are labeled (Full data can be downloaded from http://caltech.wormbase.org/pub/wormbase/datasets-published/packer2019/). The colors represent the broad cell class to which the cell type has been assigned. The size of the point is proportional to the log2 of the numbers of cells in the dataset of that cell type.
