WormBase Gene ID  ? WBGene00021929 Gene Name  dcap-1
Sequence Name  ? Y55F3AM.12 Brief Description  dcap-1 encodes an ortholog of budding yeast Dcp1p and human DCP1A(OMIM:607010) and DCP1B (OMIM:609843); DCAP-1 is expressed in diversetissues, including muscle, intestine, neurons, and the reproductivesystem; DCAP-1 is colocalized with CAR-1 to P granules, and with AIN-1and CAR-1 to discrete cytoplasmic foci that resemble P bodies; althoughit is orthologous to mRNA decapping complex subunits, DCAP-1 itselfshows no decapping activity in vitro on either m7GpppG- orm2,2,7GpppG-capped RNAs; however, dcap-1(RNAi) shows weak syntheticlethality with dcap-2(RNAi), suggesting that DCAP-1 and DCAP-2 arepartially redundant in vivo.
Organism  Caenorhabditis elegans Automated Description  Predicted to enable mRNA binding activity. Involved in several processes, including determination of adult lifespan; nematode larval development; and response to stress. Located in P granule and P-body. Expressed in head; motor neurons; neurons; tail; and touch receptor neurons. Is an ortholog of human DCP1A (decapping mRNA 1A) and DCP1B (decapping mRNA 1B).
Biotype  SO:0001217 Genetic Position  IV :-22.4137 ±0.043406

Maternal class (M): genes that are called present in at least one of the three PC6 replicates. A modified Welch F statistic was used for ANOVA. For each gene, regressed error estimates were substituted for observed error estimates. The substitution is justified by the lack of consistency among the most and least variable genes at each time point. Regressed error estimates were abundance-dependent pooled error estimates that represented a median error estimate from a window of genes of similar abundance to the gene of interest. A randomization test was used to compute the probability Pg of the observed F statistic for gene g under the null hypothesis that developmental time had no effect on expression. P-values were not corrected for multiple testing. [cgc5767]:expression_class_M

Genes that showed expression levels higher than the corresponding reference sample (L2 all cell reference). A Mann-Whitney U test with an empirical background model and FDR correction for multiple testing was used to detect expressed transcripts (Benjamini and Hochberg 1995). Genes and TARs with an FDR <= 0.05 were reported as expressed above background. Authors detected differentially expressed transcripts using a method based on linear models. Genes and TARs were called differentially expressed if the FDR was <= 0.05 and the fold change (FC) >= 2.0. To more strictly correct for potential false-positives resulting from multiple sample comparisons, authors divided individual FDR estimates by the number of samplesor sample comparisons, respectively. This resulted in an adjusted FDR of 1.3 * 0.0001 for expression above background and of 7.4 * 0.0001 for differential expression. Authors called genes selectively enriched in a given tissue if they met the following requirements: (1) enriched expression in a given tissue (FDR <= 0.05 and FC >= 2.0), (2) fold change versus reference among the upper 40% of the positive FC range observed for this gene across all tissues, and (3) fold-change entropy among the lower 40% of the distribution observed for all genes.     Expr11898   Worms expressing the Pdcap-1::gfp reporter displayed a smooth and diffused fluorescent signal throughout the cytoplasm of most cells.
Expr11899   Worms bearing the dcap-1::gfp fusion showed an ubiquitous expression pattern with occasional cytoplasmic puncta, varying among the worms.
Strain: BC13983 [Y55F3AM.12::gfp] transcriptional fusion. PCR products were amplified using primer A: 5' [GACGCTTTGATTGTTTAGATTTGA] 3' and primer B 5' [CGTCGCTGATACTGGTTCCT] 3'. New Symbol: tia-1. Picture: Fig. 1, Fig S1.   Expr8554   All P-body and stress granule GFP fusions localized to cytoplasmic foci. Some fusions appeared almost exclusively in foci (DCAP-1 and PATR-1), whereas others were also detected diffusely in the cytoplasm (CGH-1, LSM-1/-3, CCF-1, PAB-1, and TIA-1). DCAP-1, CGH-1, PATR-1, and CCF-1 fusions formed foci in both somatic and germline blastomeres. In contrast, LSM-1 and LSM-3 fusions only localized to foci in somatic blastomeres, and PAB-1 and TIA-1 fusions only localized to foci in germline blastomeres. In contrast, exosome GFP fusions were diffusely distributed in the cytoplasm and nuclei in all blastomeres and did not form distinct foci.
Expr3648   The punctate cytoplasmic pattern of AIN-1::GFP resembles that of the P bodies. By using an anti-DCAP-2 antibody, DCAP-2 has been localized to cytoplasmic foci in C. elegans embryos that resembled P bodies (Richard Davis, personal communication). DCAP-2 is the mammalian and yeast homolog of subunit Dcp2 of decapping enzymes that localizes at P bodies. Staining using this antibody and an anti-GFP antibody indicated that AIN-1 was colocalized with DCAP-2 in the embryos. AIN-1::GFP was also observed to be colocalized with a FLAG-tagged DCAP-1 protein at cytoplasmic foci in C. elegans larvae. DCAP-1 is a homolog of mammalian and yeast Dcp1 that are also a subunit of decapping enzymes localized at P bodies.
