WormBase Gene ID  ? WBGene00003886 Gene Name  osm-6
Sequence Name  ? R31.3 Brief Description  osm-6 encodes a novel protein that is orthologous to the IFT52 component of the intraflagellar transport particle; osm-6 activity is required cell autonomously for proper sensory cilium structure and thus for normal ciliated sensory neuron function; OSM-6 is a component of the intraflagellar transport particle subcomplex B, and OSM-6::GFP reporter fusions are expressed exclusively in ciliated sensory neurons.
Organism  Caenorhabditis elegans Automated Description  Involved in several processes, including cellular response to salt; non-motile cilium assembly; and sensory perception of mechanical stimulus. Located in cilium; cytoplasm; and neuronal cell body. Expressed in neurons. Human ortholog(s) of this gene implicated in asphyxiating thoracic dystrophy. Is an ortholog of human IFT52 (intraflagellar transport 52).
Biotype  SO:0001217 Genetic Position  V :3.54233 ±0.000979
Length (nt)  ? 2826

Transcripts of coding genes that showed significantly decreased expression in muscle. DESeq2 (version 1.24.0). Transcripts with a false-discovery rate adjusted p-value less than 0.05 were considered significantly differentially expressed. WBPaper00062325:muscle_depleted_coding-RNA

Genes that showed expression levels higher than the corresponding reference sample (embryonic 24hr reference). A Mann-Whitney U test with an empirical background model and FDR correction for multiple testing was used to detect expressed transcripts (Benjamini and Hochberg 1995). Genes and TARs with an FDR <= 0.05 were reported as expressed above background. Authors detected differentially expressed transcripts using a method based on linear models. Genes and TARs were called differentially expressed if the FDR was <= 0.05 and the fold change (FC) >= 2.0. To more strictly correct for potential false-positives resulting from multiple sample comparisons, authors divided individual FDR estimates by the number of samplesor sample comparisons, respectively. This resulted in an adjusted FDR of 1.3 * 0.0001 for expression above background and of 7.4 * 0.0001 for differential expression. Authors called genes selectively enriched in a given tissue if they met the following requirements: (1) enriched expression in a given tissue (FDR <= 0.05 and FC >= 2.0), (2) fold change versus reference among the upper 40% of the positive FC range observed for this gene across all tissues, and (3) fold-change entropy among the lower 40% of the distribution observed for all genes. Clone: pUL#JS1G1   Expr7645 Weak expression is seen in the nerve cell bodies and nerve cell processes of the head and tail region. Expression is seen from mid stage embryos onwards.
Included upstream of gfp in the fusion gene was 5.1 kb of osm-6 genomic sequence, extending from 2.4 kb upstream of the 5' end of the osm-6 cDNA through the first 10 exons and all introns into the last exon, such that only the last 13 codons, including the translational stop, of OSM-6 were lacking. Transgenic Marker: rol-6(su1006) late embryo(author) = fully-elongated embryo(curator).   Expr507 Expressed in 56 cells in the hermaphrodite: the 24 amphid neurons, twelve inner labial neurons, six outer labial neurons, four CEP cells, two ADE cells, four phasmid cells in all stages from late embryo through adult; AQR and PQR at L1 through adult PDE cells at L2 through adult. Additional expression in male-specific cells: CEM cells and male tail sensory cells.
Picture: Figure 3.   Expr8093   GFP fusion protein was localized throughout the AWB cilia.
Promoter::GFP fusion constructs of variable lengths were generated using PCR amplification followed by subsequent cloning into expression vectors.   Expr10117 Expressed in both the amphid and tail ciliated sensory neurons. The 60 C. elegans CSNs present in an adult hermaphrodite worm were divided into 5 subgroups or anatomical regions including, neurons that reside in the amphids (region 1 = 24 CSNs: AWAL/R, AWBL/R, AWCL/R, AFDL/R, ASEL/R, ADFL/R, ASGL/R, ASHL/R, ASIL/R, ASJLR, ASKL/R, ADLL/R) or in the tail (region 2 = 5 CSNs: PHAL/R, PHBL/R, PQR), neurons surrounding the anterior bulb (region 3 = 24 CSNs: BAGL/ R, CEPVL/R, CEPDL/R, IL1L/R, IL2L/R, IL1VL/R, IL2VL/R, IL1DL/R, IL2DL/R, OLLL/R, OLQVL/R, OLQDL/R) or the posterior bulb (region 4 = 5 CSNs: ADEL/R, FLPL/R, AQR) of the pharynx, and neurons in the midbody region of the worm (region 5=2 CSNs: PDEL/R). osm-6 was detected in all 5 regions.
Expr3028 KAP::GFP, OSM-1::GFP, and OSM-6::GFP fusion proteins are expressed in amphid and phasmid chemosensory neurons. They appear to concentrate at the base of the transition zones, which correspond to the basal bodies of motile and sensory cilia. These fusion proteins are seen to emerge from the transition zones as small fluorescent particles that move in a bidirectional fashion along the sensory cilia, displaying anterograde movement from the transition zone toward the tip of the ciliary axoneme and retrograde movement from the cilium tip back toward the transition zone.
Expr12033 The cilium structural gene osm-6 is expressed in both HOA (P10.pppa) and HOB (P10.ppap).
Expr13933 The transcriptional reporter using the promoter region of the cilia specific gene osm-6 (Posm-6) is expressed in both the PQR as well as the URX neuron.
