WormBase Gene ID  ? WBGene00004053 Gene Name  tank-1
Sequence Name  ? ZK1005.1 Brief Description  tank-1 (formerly known as pme-5) encodes a protein containing N-terminal ankyrin repeats and C-terminal poly (ADP-ribose) polymerase (PARP) regulatory and catalytic domains that is orthologous to human tankyrase 1 (TNKS; OMIM:603303) and tankyrase 2 (TNKS2; OMIM: 607128), but structurally more similar to the ADPRT-3 tankyrase of Dictyostelium discoideum; TANK-1 also contains a lysine-rich region, a WGR domain, which may mediate DNA interaction, and a coil-coiled region; X-ray-induced apoptosis of germ cells is slightly increased in tank-1(RNAi) animals; DNA damage via ionizing radiation induces tank-1 expression, and this induction absolutely requires HUS-1; TANK-1 is ubiquitously expressed throughout the life cycle, including in the germ line, and localizes to the nucleus.
Organism  Caenorhabditis elegans Automated Description  Predicted to enable NAD+ ADP-ribosyltransferase activity and NAD+-protein ADP-ribosyltransferase activity. Involved in intrinsic apoptotic signaling pathway in response to DNA damage; negative regulation of programmed cell death; and response to gamma radiation. Located in nucleus.
Biotype  SO:0001217 Genetic Position  V :-19.844 ±0.004452
Length (nt)  ? 9293

Transcripts that showed significantly increased expression in L1 neural cells comparing to in adult neural cells. DESeq2 (v1.18.1) fold change > 2, P-adj<0.05, using BenjaminiHochberg correction. WBPaper00060811:L1_vs_adult_upregulated_neural
Transcripts of coding genes that showed significantly decreased expression in muscle. DESeq2 (version 1.24.0). Transcripts with a false-discovery rate adjusted p-value less than 0.05 were considered significantly differentially expressed. WBPaper00062325:muscle_depleted_coding-RNA
Transcripts expressed in neuronal cells, by analyzingfluorescence-activated cell sorted (FACS) neurons. DESeq. False discovry rate (FDR) < 0.1. WBPaper00048988:neuron_expressed
Genes that showed expression levels higher than the corresponding reference sample (embryonic 24hr reference). A Mann-Whitney U test with an empirical background model and FDR correction for multiple testing was used to detect expressed transcripts (Benjamini and Hochberg 1995). Genes and TARs with an FDR <= 0.05 were reported as expressed above background. Authors detected differentially expressed transcripts using a method based on linear models. Genes and TARs were called differentially expressed if the FDR was <= 0.05 and the fold change (FC) >= 2.0. To more strictly correct for potential false-positives resulting from multiple sample comparisons, authors divided individual FDR estimates by the number of samplesor sample comparisons, respectively. This resulted in an adjusted FDR of 1.3 * 0.0001 for expression above background and of 7.4 * 0.0001 for differential expression. Authors called genes selectively enriched in a given tissue if they met the following requirements: (1) enriched expression in a given tissue (FDR <= 0.05 and FC >= 2.0), (2) fold change versus reference among the upper 40% of the positive FC range observed for this gene across all tissues, and (3) fold-change entropy among the lower 40% of the distribution observed for all genes.   mRNAs that showed differential expression in activated AAK-2(uthIs202[aak-2 (intron 1)::aak-2(aa1-aa321)::Tomato::unc-54 3'UTR + rol-6(su1006)]), activiated CRTC-1 (uthEx222[crtc-1p::crtc-1 cDNA (S76S, S179A)::tdTomato::unc-54 3'UTR + rol-6(su1006)]), or AAK-2;CRTC-1 comparing to in N2. Genes were tested for differential expression between each mutant strain and wild-type using edgeRs glm method. Briefly, negative binomial models were fitted and dispersion estimates obtained. Picture: Fig 4, Fig 6.   Expr8972   Fluorescent signal specific for PME-5 also reveals its presence in the pachytene and diplotene zones where chromatin further condenses into chromosomes. Oocytes at the diakinesis stage, where condensation of chromosome is at its highest, showed the presence of PME-5 on chromosomes. Association of PME-5 with chromosomes is not restricted to the female germ cells. Indeed, nuclei of sperms resident in the spermatheca were labeled with anti-PME-5 antibody. The association of PME-5 with chromatin was observed in a special structure called anaphase bridges. Anaphase bridges are formed during the separation of the two chromosomes. This structure occurs predominantly when dicentric chromosomes are present resulting from the fusion of two telomere-free fragments. This observation further strengthened the possible interaction between PME-5 and chromatin. One of the most striking features of immunolocalization of PME-5 is its co-localization with chromosomes. This is seen during the early embryogenesis where cell divisions occur for the establishment of the different axis. During the second round division in the early embryos, just prior the cell division of the two founder blastomeres (AB and P1), the condensing chromosomes are clearly labeled with anti-PME-5 antibody. The distal part of the gonad comprises the mitotic zone, located between the distal tip cell and the transition zone. The germ cells in the transition zone enter the leptotene and zygotene phases, which are the early phases of meiotic prophase, and the DNA in the nucleus starts to condense. The nuclei of these germ cells were visible when probed for the presence of PME-5, suggesting an association with chromatin rather than condensed chromosomes at this moment in oogenesis.
