Subcellular Localization  HYLS-1 localized to punctate structures in amphid and labial/cephalic neurons. This punctate staining was absent in hyls-1(D) worms and was fully restored. HYLS-1 puncta were closely apposed to the short stretches of NPH-4:YFP, a marker for the ciliary transition zone, indicating that HYLS-1 localizes to a discrete structure at the base of cilia. This result was confirmed using CHE-GFP, an IFT component that localizes to the transition zone and along the ciliary axoneme. Localization to the base of the cilium was also observed in phasmid neurons. Thus, HYLS-1 localizes to a discrete structure at the base of the mature cilium.
