WormMine: ExpressionPattern Expr3260

• Pattern: Both the sth-1::lacZ fusion gene pS3434Z and the sth-1::gfp fusion gene pS3434G were expressed in germ line cells around the vulva in the L4 stage and the spermatheca in adult. Both the fusion genes were expressed in the hermaphrodite after the L4 stage to the adult stage; however, they were not expressed in male gonads. Spermathecal localization of sth-1 transcripts was confirmed in the same tissues by in situ hybridization. The promoter deletion construct, pS1292G was also expressed in spermatheca.

• Remark: The additional promoter deletion construct pS672Z was not expressed in any stage in hermaphrodites. These results indicate that the 0.7 - 1.3 kb upstream region of sth-1, represented as SE (spermathecal expression), was required for the expression of sth-1 in spermatheca.
• Subcellular Localization: pS3434Z and pS3434G was expressed in the nuclei and cytoplasm, respectively. The sth-1::gfp fusion gene pSTL1292G, which contains the full-length sth-1 coding regions and the gfp reporter gene, was expressed in spermatheca associated with the cell membrane.
