WormBase Gene ID  ? WBGene00002034 Gene Name  htp-3
Sequence Name  ? F57C9.5 Brief Description  htp-3 encodes a HORMA domain-containing protein that is one of three C. elegans HIM-3 paralogs; during meiosis, htp-3 activity is required for homologous chromosome alignment and synapsis, and for meiotic double strand break formation; immunoprecipitation experiments indicate that HTP-3 interacts in vivo with complexes containing MRE-11 and RAD-50, and HIM-3; HTP-3 is expressed in the germline where it localizes to chromatin in premeiotic nuclei and, with HIM-3, to the axes, or cores, of meiotic chromosomes; HTP-3 localization to premeiotic chromatin and meiotic chromosomes is dependent upon scc-3 activity, while in turn HIM-3 localization to meiotic chromosomal axes, and SYP-1 localization to the synaptonemal complex, is dependent upon HTP-3 activity.
Organism  Caenorhabditis elegans Automated Description  Involved in homologous chromosome pairing at meiosis and meiotic DNA double-strand break formation. Located in lateral element. Part of chromatin. Expressed in Z2; Z3; embryonic cell; germ line; and neurons.
Biotype  SO:0001217 Genetic Position  I :-0.718461 ±0.003661
Length (nt)  ? 2720

Genes that were upregulated in lin-15B(n744). For each gene in each microarray hybridization experiment, the ratio of RNA levels from the two samples was transformed into a log2 value and the mean log2 ratio was calculated. The log2 ratios were normalized by print-tip Loess normalization (Dudoit and Yang, 2002). All genes with a false discovery rate of <= 5% (q <= 0.05) (Storey and Tibshirani, 2003) and a mean fold-change ratio of >= 1.5 were selected for further analysis.   Maternal class (M): genes that are called present in at least one of the three PC6 replicates. A modified Welch F statistic was used for ANOVA. For each gene, regressed error estimates were substituted for observed error estimates. The substitution is justified by the lack of consistency among the most and least variable genes at each time point. Regressed error estimates were abundance-dependent pooled error estimates that represented a median error estimate from a window of genes of similar abundance to the gene of interest. A randomization test was used to compute the probability Pg of the observed F statistic for gene g under the null hypothesis that developmental time had no effect on expression. P-values were not corrected for multiple testing. [cgc5767]:expression_class_M
Dietary restriction Transcripts that showed significantly decreased expression after N2 animals were under dietary restriction (DR, OP50 OD = 0.1) from 3-day post L4 till 6-day post L4 adult hermaphrodite stage, comparing to under ad libtum (AL, OP50 OD = 3) condition. Bioconductor package edgeR, p < 0.05. WBPaper00056443:DietaryRestriction_downregulated
Strictly maternal class (SM): genes that are the subset of maternal genes that are not also classified as embryonic. [cgc5767]:expression_class_SM

Picture: Fig 8A (wild type panel).   Expr8709   HTP-3 and SYP-1 colocalize in pachytene nuclei of wild-type animals. Cruciform HTP-3 staining is observed on diakinesis bivalents.
Picture: Figure 1A.   Expr8000   HTP-3 localized to all nuclei within the germline, both those proliferating mitotically and those engaged in the meiotic cell cycle. In premeiotic nuclei, HTP-3 staining assumed a diffuse pattern that overlapped the DAPI-stained chromatin in interphase nuclei but was not detectable on the condensed chromatin of M-phase nuclei. Upon entry into meiotic prophase, HTP-3 localized to chromosome axes, and colocalized with HIM-3 throughout meiotic prophase.
Expr11541 Immunostaining for HTP-3 is easily detected in embryonic Z2/Z3. The protein appears to be maternally loaded in early embryos, to diminish significantly by the P4 stage, and then to turn on in Z2/Z3.
