Picture: Fig 3.   Expr8697 klf-3::gfp expression first appeared in the early larvae which were still enclosed in the eggshell of the embryo. During larval development, gfp fluorescence was frequently observed in the intestinal cells of developing larvae, young adults, egg-laying hermaphrodites, and male worm. Gfp fluorescence was very strong and persisted even in very old adult worms. This pattern was consistently seen in all three transgenic lines.  
Picture: Fig 2.   Expr8696 klf-3/mua-1 is expressed throughout the lifespan of the worm but in varying abundances. The total amount of klf-3 transcripts was lower in embryos and higher in developing larvae. The peak amount was seen in L1 larvae. After L1 growth, the klf-3 level dropped slightly in the L2, L3, and L4 stages  
    Expr9597 The klf-3b isoform, similar to klf-3a, is also expressed in the intestine during all larval and adult stages. KLF3 protein expression is absent from both the mitochondria and the peroxisomes of intestinal cells (data not shown).  
    Expr9823 Nuclear-localized GFP was observed from late embryo to mature adult. Intestinal expression and expression in hypodermal nuclei along the sides of the head was seen from the L1 stage, and continues throughout development although much weaker in the adult. Several cells in the tail, that may be rectal cells, and in the adult vulva also expressed GFP. There was also much weaker GFP expression along adult body, in the hypodermis or body wall muscle.  
    Expr11008 In wild-type worm, KLF-3 protein was undetectable in embryos. However, KLF-3 expression was evident in all larval stages, as well as in adult. Microscopic observation of animals at larval and adult stages showed that level of KLF-3 expression was constant in each developmental stage. Throughout worm development, the anti-KLF-3 antibodies detected KLF-3 protein in the intestine, where it was most abundant. We also observed KLF-3 protein in both arms of the germline.  
    Expr9810 GFP expression was observed from the comma stage embryo. The GFP may be in the P cell lineages from late embryogenesis and through the L1 stage but this seems to disappear at the L2 stage. A few cells in the body wall expressed GFP in older larvae and adults; this may be muscle but was very weak and may be hypodermis in the head. There may also be GFP expression in vulval and rectal muscle in the L4 and young adult. No intestinal expression of GFP was detected.  
    Expr9814 Intestinal GFP expression was observed from the L1 to the adult. The GFP was largely nuclear-localized but with some cytoplasmic fluorescence. GFP may have been present in the P cell lineages in the L1, in the hypodermis either side of the head, and possibly some body wall muscle in the head. Vulval and rectal GFP expression may be in muscle cells. UL4026 showed stronger GFP expression than UL4025 or UL4008, but this was still weaker than observed for strains expressing N-terminal reporter fusions for klf-3, generated by a similar strategy (see UL4047 and UL4006).
