    Expr3696 Fluorescence was observed in epithelial cells that synthesize cuticle. Expressed in the hypodermis, including the major body syncytium, hyp7, and hypodermal cells in the head and tail. A pulse of fluorescence was observed in the hypodermis prior to molting. Expression in the hypodermis was observed starting 63 +/- 2% of the way through each larval stage. Expression of the gfp fusion genes was never detected in the hypodermis of gravid adults that no longer molt.  
New Anatomy_term: embryo epidermal cell Picture: Figure 4A.   Expr9014 The mlt-10p::gfp-pest fusion gene was expressed in the major body hypodermal syncytium (Hyp7), the dorsal and ventral ridges of the hypodermis, hypodermal cells in the head and tail, and the pharyngeal myoepithelium, but not the lateral seam cells. In L4-stage animals cultured at 25 centigrade, GFP was first detected in the anterior hypodermis about 3.5 hours prior to ecdysis. The fluorescence spread throughout Hyp7 and intensified for about three hours. The fluorescence dissipated at the end of lethargus, and was barely detectable 1 hour after ecdysis. GFP was also expressed in epidermal cells of pretzel-stage embryos, which synthesize the cuticle for the first larval stage.  
Picture: Fig 5.   Expr9015   During the process of molting, the MLT-10::mCherry and GFP::MLT-10 fusion proteins were detected in vesicle-like objects near the apical surface of the epidermis. Cherry was also readily detected in other structures, whereas GFP was not, possibly due to proteolytic processing of the N-terminal fusion protein. In young adults, the MLT-10::mCherry fusion protein was also detected in coelomocytes. Cherry was detected in the coelomocytes of approximately 22% (n=176) of aaaEx16[mlt-10::mCherry] animals. The MLT-10::mCherry fusion protein was also detected in strands and loops near the surface of transgenic animals. These structures were observed in 27% (n=123) of mixed-stage larvae, but were most prominent in animals completing the fourth molt. The strands ranged from 5 to 30 microns in length and appeared to be positioned above the hypodermis. The formal possibility that these structures result from the non-specific aggregation of mCherry cannot be eliminated at this time.
