Subcellular Localization  Antibody staining against the vesicular acetylcholine transporters UNC-17 and epitope-tagged acetylcholine receptors (UNC-29 and UNC-38) showed that UNC-122 clusters precisely oppose UNC-17 clusters, consistent with the localization of UNC-122 to muscle endplates. UNC-122 clusters do not precisely overlap but rather interdigitate with clusters of UNC-38 AChR or UNC-29; the relatively large size of muscle endplates suggests that UNC-122 and UNC-38 may localize to distinct subdomains within muscle end-plates. Together with the demonstration of a muscular focus of action of unc-122, authors conclude from these localization experiments that UNC-122 localizes to the postsynaptic side of NMJs. Epitope-tagged UNC-122 shows a punctate localization along the ventral nerve cord (VNC). These sites do not coincide with muscle-hypodermis attachment sites, as observed by antibody costaining against an attachment marker protein.
