WormBase Gene ID  ? WBGene00003245 Gene Name  mig-13
Sequence Name  ? F43C9.4 Brief Description  mig-13 encodes a novel single-pass transmembrane protein with a N-terminal signal sequence and two motifs that have been implicated in protein-protein interactions: a CUB domain and a LDL receptor repeat; mig-13 affects the migration and thus the positioning of the Q neuroblasts and their descendants and also the anterior migration of the BDU neuron and may be a component of a global anterior/posterior migration system; a mig-13-gfp translational fusion protein is expressed in embryonic pharyngeal, hypodermal and anterior body neuronal precursors as well as in the cell bodies and axons of a number of neurons in the retrovascular ganglion and the ventral cord; mig-13 localizes to the plasma membrane; mig-13 expression along the A/P axis is restricted by hox gene activity.

Transcripts that showed significantly increased expression in L1 neural cells comparing to in adult neural cells. DESeq2 (v1.18.1) fold change > 2, P-adj<0.05, using BenjaminiHochberg correction. WBPaper00060811:L1_vs_adult_upregulated_neural
Transcripts expressed in neuronal cells, by analyzingfluorescence-activated cell sorted (FACS) neurons. DESeq. False discovry rate (FDR) < 0.1. WBPaper00048988:neuron_expressed
Genes significantly enriched (> 2x, FDR < 5%) in a particular cell-type versus a reference sample of all cells at the same stage. A Mann-Whitney U test with an empirical background model and FDR correction for multiple testing was used to detect expressed transcripts (Benjamini and Hochberg 1995). Genes and TARs with an FDR <= 0.05 were reported as expressed above background. Authors detected differentially expressed transcripts using a method based on linear models. Genes and TARs were called differentially expressed if the FDR was <= 0.05 and the fold change (FC) >= 2.0. To more strictly correct for potential false-positives resulting from multiple sample comparisons, authors divided individual FDR estimates by the number of samplesor sample comparisons, respectively. This resulted in an adjusted FDR of 1.3 * 0.0001 for expression above background and of 7.4 * 0.0001 for differential expression. Authors called genes selectively enriched in a given tissue if they met the following requirements: (1) enriched expression in a given tissue (FDR <= 0.05 and FC >= 2.0), (2) fold change versus reference among the upper 40% of the positive FC range observed for this gene across all tissues, and (3) fold-change entropy among the lower 40% of the distribution observed for all genes. WBPaper00037950:A-class-motor-neurons_larva_enriched

Expr4863 A-class motor neuron: enriched in embryo (3.0) and larva (2.8). Neuronal expression include: Bright in anterior ventral cord neurons, weak and mosaic in posterior ventral cord. Also expressed in other cells: Pharyngeal/intestinal valves, hypodermis. Pan-neuronal: expressed in embryo; enriched in larva (1.8).

Expr15167 Using an MIG-13::GFP knockin strain, we confirmed an even distribution of MIG-13 in migrating QR.ap (AQR) cells. MIG-13 evenly distributed to the entire plasma membrane.
Expr11079 MIG-13::GFP fluorescence was not observed in QL.x. However, MIG-13::GFP is expressed in the migrating QR.x, and its GFP fluorescence is present on the cell surface. Time-lapse imaging revealed dynamic changes of mig-13 expression during migration: MIG-13::GFP was detectable at the initial stage of QR.x migration; MIG-13::GFP gradually increased its fluorescence during migration and reached its maximum level at the end of migration. MIG-13::GFP fluorescence in QR.a/ap is stronger than that observed in QR.p/pa. The presence of MIG-13::GFP in QR.x was confirmed using an independent transgenic integrant (muIs42).
Expr1335 mig-13::GFP was first seen during embryogenesis in pharyngeal, hypodermal, and neuronal precursors located in the anterior body region. At hatching, mig-13::GFP was expressed in the pharyngeal-intestinal valve cells, as well as in the cell bodies and axons of a number of neurons in the retrovesicular ganglion and the ventral cord. Expression appeared to be restricted to cells in the anterior half of the body with the exception of DA9, which is located in the preanal ganglion. In later larval and adult stages, the same cells continued to express mig-13::GFP, although additional staining was seen in the postembryonically derived ventral cord neurons located in the anterior half of the animal. Significantly, no expression was seen in the migrating QR descendants. MIG-13 appeared to be localized to the plasma membrane of all mig-13 expressing cells.
    Expr13482
Expr9185 Pmig-13::mig-13::gfp in the tail is predominantly expressed in DA9 during larval stages 1 through 4 (L1-L4).

Expr2013561 Single cell embryonic expression. Only cell types with an expression fraction of greater 0.2 of the maximum expressed fraction are labeled (Full data can be downloaded from http://caltech.wormbase.org/pub/wormbase/datasets-published/packer2019/). The colors represent the broad cell class to which the cell type has been assigned. The size of the point is proportional to the log2 of the numbers of cells in the dataset of that cell type. Interactive visualizations are available as a web app (https://cello.shinyapps.io/celegans/) and can also be installed as an R package (https://github.com/qinzhu/VisCello.celegans).
Expr1170024 Time-lapse fluorescence microscopy was performed, including DIC for morphology. Gene expression patterns were summarized in 4 manners: Average over time, Average over time and at different positions along the anterior-posterior (AP) axis, a voxelized representation over time, and on individual cells overlaid from a reference coordinate dataset (https://doi.org/10.1016/j.ydbio.2009.06.014). The analysis was done with a pipeline based on the multi-purpose image analysis software Endrov (https://doi.org/10.1038/nmeth.2478), which further is needed to browse the raw recording data. Thumbnail movies were also generated, using maximum Z projection for the 3D fluorescence channel.
