Y69A2AR.2 Brief Description  ric-8 encodes a receptor-independent G alpha guanine nucleotide exchange factor (GEF); ric-8 activity is required for regulating heterotrimeric G protein signaling pathways that control asymmetric cell division in the embryo as well as synaptic transmission in the nervous system; in the 1-cell embryo, RIC-8 functions as a GEF for GOA-1 thereby regulating spindle morphology, position, and consequently, the first asymmetric cell division; in the nervous system, RIC-8 likely functions to regulate activity of both the EGL-30 and GSA-1 pathways to regulate neurotransmitter secretion and thus, coordinated locomotion; RIC-8 also functions, with AGS-3, to activate GOA-1/Galpha0 in the ASH chemosensory neurons to modulate behavior after food deprivation; in vitro, RIC-8 preferentially binds GOA-1-GDP and increases the rate of GOA-1 GTP binding; in the early embryo, RIC-8 localizes to the cellular cortex, junctions between cells, asters of the mitotic spindle, the central spindle (kinetochore microtubules), the nuclear envelope, and chromatin; RIC-8's localization to the cortex and asters mirrors that of GOA-1, while localization to the cortex and central spindle is similar to that of the GOA-1 regulators GPR-1/2, whose subcellular localization is RIC-8-dependent; in later stage animals, RIC-8 is seen in head and tail ganglia, amphid processes, and the ventral nerve cord, as well as in larval nonneuronal cells, such as germline nuclei, where it localizes near the nuclear membrane.

The failure to detect cortical fluorescence at the periphery of embryos or the zygote may simply be due to the fact that only a single cell cortex is present, rather than the two opposed cortices present at junctions between cells.   Expr3109   For RIC-8::GFP, faint cortical localization was seen at the junctions between cells. RIC-8::GFP showed an additional, dynamic subcellular localization. It was excluded from interphase nuclei and became localized around what appeared to be kinetochore microtubules at the onset of mitosis, becoming more diffuse during anaphase. This pattern was observed beginning with division of the zygote and repeated in each subsequent cell division.
With the exception of the staining at the nuclear envelope and around the chromatin, the RIC-8 staining is reduced in ric-8(md1909);ric-8(RNAi) but not completely abolished, consistent with a partial RNAi effect. All staining patterns are absent in a preadsorbed antibody preparation, confirming the specificity of the antibodies. Two additional lines of evidence suggest that the nuclear envelope staining is specific. This localization was also observed with a different antibody generated by Miller and Rand, and GFP::RIC-8 embryos show the same localization pattern.   Expr3393   The subcellular localization of RIC-8 is very similar to the localization of GOA-1, for example at the cortex and on the asters of the mitotic spindle. In addition, RIC-8 was found to be localized on the central spindle, as GPR-1/2, at the nuclear envelope and around the chromatin.
    Expr2033583 Single cell embryonic expression.     Expr3105   The bulk of the signal is cytoplasmic in one-cell stage embryos, with a slight enrichment in the vicinity of microtubule asters. In addition, some signal is present at the cell cortex, something that is more apparent in two- or four-cell stage embryos. The cytoplasmic and cortical distributions correspond to bona fide RIC-8 because they are significantly diminished in ric-8(RNAi) embryos. RIC-8 antibodies also decorate the nuclear periphery, but this signal does not appear specific because it is still present in ric-8(RNAi) embryos, a conclusion also reached when examining neurons. The distribution of RIC-8 is not altered in ric-8(md1909) or ric-8(md303) mutant embryos, nor in goa-1/gpa-16(RNAi) or gpr-1/2(RNAi) embryos. Taken together, these observations indicate that the bulk of RIC-8 in early embryos is cytoplasmic, although a fraction of the protein is found at the cell cortex.
