The immunostaining patterns described here were not present in putative null unc-83 mutants and thus are specific for the UNC-83 protein. UNC-83 co-localized with two other proteins of the nuclear envelope, UNC-84 and lamin. UNC-83 has a more punctate staining pattern than Lamin. It also appears that UNC-83 is more punctate that UNC-84. This may simply be due to the fact that UNC-84 was detected as a GFP fusion protein that is likely to be overexpressed. unc-83(ku18) mutants, which have nuclear migration defects in the P cells and the intestinal cells but not in hyp7 precursors, showed immunostaining for UNC-83 in the hyp7 nuclei but not in the P cell nuclei or intestinal nuclei.   Expr1789 UNC-83 was not detected at the nuclear envelope of migrating pronuclei. UNC-83 was first detected at the nuclear envelopes of migrating embryonic hyp7 nuclei. Later, at the bean stage of embryonic development, UNC-83 was localized to the nuclear envelopes of hyp7 cells, P cells and intestinal cells. By the comma stage, UNC-83 was detected in these cells as well as in several additional cells in the pharynx. In late stages of wild-type embryos, and in larvae and adults, UNC-83 was detected on nuclei in a wide variety of cells, including several cells around the pharynx and in the uterus. Antibodies against UNC-83 stained the nuclear envelopes of migratory nuclei and several additional nuclei.
