WormBase Gene ID  ? WBGene00006957 Gene Name  wsp-1
Sequence Name  ? C07G1.4 Brief Description  wsp-1 encodes the C. elegans WASP ortholog; when mutated, human WASP leads to Wiskott-Aldrich syndrome (OMIM:301000); wsp-1 is required for hypodermal cell migration during morphogenesis (ventral enclosure); in a yeast two-hybrid assay WSP-1 physically interacts with WIP-1, the C. elegans WASP-interacting protein ortholog; WSP-1 and WIP-1 are mutually required for normal protein levels; WSP-1 is found in migrating hypodermal cells during ventral enclosure.
Organism  Caenorhabditis elegans Automated Description  Enables cytoskeletal regulatory protein binding activity and small GTPase binding activity. Involved in several processes, including embryonic morphogenesis; positive regulation of egg-laying behavior; and regulation of cellular component organization. Located in cell leading edge; cortical actin cytoskeleton; and presynaptic periactive zone. Expressed in Q cell; anchor cell; apoptotic cell; epithelial cell; and hypodermis. Is an ortholog of human WASL (WASP like actin nucleation promoting factor).
Biotype  SO:0001217 Genetic Position  IV :3.65746 ±0.002507
Length (nt)  ? 10048

Regulated By Treatment Description Algorithm Primary Identifier
Transcripts expressed in neuronal cells, by analyzingfluorescence-activated cell sorted (FACS) neurons. DESeq. False discovry rate (FDR) < 0.1. WBPaper00048988:neuron_expressed
Genes that showed expression levels higher than the corresponding reference sample (embryonic 24hr reference). A Mann-Whitney U test with an empirical background model and FDR correction for multiple testing was used to detect expressed transcripts (Benjamini and Hochberg 1995). Genes and TARs with an FDR <= 0.05 were reported as expressed above background. Authors detected differentially expressed transcripts using a method based on linear models. Genes and TARs were called differentially expressed if the FDR was <= 0.05 and the fold change (FC) >= 2.0. To more strictly correct for potential false-positives resulting from multiple sample comparisons, authors divided individual FDR estimates by the number of samplesor sample comparisons, respectively. This resulted in an adjusted FDR of 1.3 * 0.0001 for expression above background and of 7.4 * 0.0001 for differential expression. Authors called genes selectively enriched in a given tissue if they met the following requirements: (1) enriched expression in a given tissue (FDR <= 0.05 and FC >= 2.0), (2) fold change versus reference among the upper 40% of the positive FC range observed for this gene across all tissues, and (3) fold-change entropy among the lower 40% of the distribution observed for all genes.   mRNAs that showed decreased expression in 1 cell mebryo comparing to in oocyte, according to RNAseq analysis. Gaussian error propagation. As cutoff for the up-regulated genes authors used log2 fold change > 1 and P < 0.05 and as cutoff for the down-regulated genes authors used log2 fold change < -1 and P < 0.05.   Maternal class (M): genes that are called present in at least one of the three PC6 replicates. A modified Welch F statistic was used for ANOVA. For each gene, regressed error estimates were substituted for observed error estimates. The substitution is justified by the lack of consistency among the most and least variable genes at each time point. Regressed error estimates were abundance-dependent pooled error estimates that represented a median error estimate from a window of genes of similar abundance to the gene of interest. A randomization test was used to compute the probability Pg of the observed F statistic for gene g under the null hypothesis that developmental time had no effect on expression. P-values were not corrected for multiple testing. [cgc5767]:expression_class_M
Genes with expression level regulated by genotype (N2 vs CB4856) and age at old adults stage (214 hours at 24 centigrade). For model 2, authors used 100 permutations to estimate the FDR threshold. Per permutation, genotypes and ages were independently randomly distributed, keeping the among-gene structure intact. Then for each spot (23,232) on the array, model 2 was tested. The obtained P-values were used to estimate a threshold for each of the explanatory factors. Authors also used a genome-wide threshold of -log10 P-value = 2, which resembles an FDR of 0.072 and 0.060 for marker and the interaction age-marker for the developing worms and FDR of 0.050 and 0.065 for marker and age-marker for the aging worms. For the physiological age effect, authors used a log10 P-value = 8 in developing worms (0.012 FDR) and -log10 P-value = 6 (0.032 FDR). WBPaper00040858:eQTL_age_regulated_aging

Strictly maternal class (SM): genes that are the subset of maternal genes that are not also classified as embryonic. Expr1032968 Tiling arrays expression graphs
Expr15180 GFP::WSP-1A is specifically localized in the socket cells but is barely detectable in the cilia or the sheath cells.
In wsp-1(RNAi) embryos, WSP-1 protein levels were reduced and localization at boundaries was diminished. Reduction of the protein level of WSP-1 was observed in almost all the RNAi-treated embryos, including those that would undergo normal embryogenesis. Anti-actin staining of wsp-1(RNAi) embryos did not significantly alter the actin skeleton compared to wild-type embryos.   Expr2466   During early embryogenesis, WSP-1 colocalized with actin at the blastomere boundary. During ventral enclosure, WSP-1 colocalized with actin at the periphery of cells, including in hypodermal cells migrating to the ventral side.
Picture: Fig 4.   Expr9050   GFP expression was observed diffused in the nerve cords but was also enriched in discrete punctate structures along the ventral nerve cord. WSP-1 localized to regions immediately adjacent to RAB-3 puncta. This region of WSP-1 localization corresponds to periactive zone localization, which is F-actin rich region that regulates synaptic growth and plasticity. Therefore, WSP-1::GFP fusion protein localizes to the pre-synaptic region of the neuromuscular junction.
Expr13968   WSP-1/N-WASP was present at the Anchor Cell invasive membrane before and after invasion, although it was also visible at cell junctions elsewhere in the vulval and uterine cells. Spots of WSP-1/N-WASP on the invasive membrane colocalized with spots of WVE-1/WAVE.
Expr12696 GFP::WSP-1 was found in punctate structures at the anchor cells (AC) invasive cell membrane that colocalized with F-actin puncta, indicating that WSP-1 localizes to AC invadopodia.
Expr13651 WASP was detected at the leading edge, and under a 1503 objective the GFP::WSP-1 signal was resolved to separate puncta, which was not previously reported in migrating cells of live animals but may represent its real behavior at the endogenous expression level. GFP::WSP-1, GFP::WVE-1, and ARX-2::GFP occupied 36%, 53%, and 89%, respectively, of the periphery of the leading edge.
Expr13400 A triple fluorescence animal that expresses ARX-2::TagRFP knock-in, GFP::WSP-1 knock-in, and the Q cell marker Plin-32::TagBFP shows that the majority of ARX-2:: TagRFP was recruited on the Q cell corpse at approximately the same time as GFP::WSP-1 and that the ARX-2::TagRFP fluorescence was largely co-localized with GFP::WSP-1 (n = 10).
