Sequence Name  ? C09B8.6 Brief Description  hsp-25 encodes a member of the small heat shock family of proteins; HSP-25 is expressed throughout development and in vitro, exhibits chaperone activity; HSP-25 localizes to: 1) dense bodies and M lines in body wall muscle, 2) the lining of the pharynx, and 3) to cell-cell junctions in the spermathecal wall; consistent with a role in myofibril organization, HSP-25 binds vinculin and alpha-actinin in vitro.
Organism  Caenorhabditis elegans Automated Description  Predicted to enable unfolded protein binding activity. Predicted to be involved in protein refolding and response to heat. Located in M band and striated muscle dense body. Expressed in body wall musculature; gonad; pharynx; and spermatheca. Human ortholog(s) of this gene implicated in Charcot-Marie-Tooth disease axonal type 2L and autosomal dominant distal hereditary motor neuronopathy 2. Is an ortholog of human HSPB8 (heat shock protein family B (small) member 8).

Maternal class (M): genes that are called present in at least one of the three PC6 replicates. A modified Welch F statistic was used for ANOVA. For each gene, regressed error estimates were substituted for observed error estimates. The substitution is justified by the lack of consistency among the most and least variable genes at each time point. Regressed error estimates were abundance-dependent pooled error estimates that represented a median error estimate from a window of genes of similar abundance to the gene of interest. A randomization test was used to compute the probability Pg of the observed F statistic for gene g under the null hypothesis that developmental time had no effect on expression. P-values were not corrected for multiple testing. [cgc5767]:expression_class_M

Strictly maternal class (SM): genes that are the subset of maternal genes that are not also classified as embryonic. When an excess of recombinant HSP25 was included in the reactions as competitor, no staining was observed, indicating that the pattern seen is specific to HSP25.   Expr1088 In both larvae and adults, strong staining was observed in body wall muscle and in the lining of the pharynx. The pharyngeal staining is localized to a subset of cells bordering the lumen. strong HSP25 staining was also observed at the junctions between cells forming the spermathecal wall. HSP25 appears to be associated with desmosomes in the spermatheca. In body wall muscle, HSP25 was localized to a series of thin lines parallel to the long axis of the muscle fiber, alternating with thicker lines consisting of discrete spots. This pattern is consistent with the localization of HSP25 to the dense bodies and M-lines of the myofibrils.
Expr15507 HSP-25 and HSP-12.1 were localized to the M-lines and dense bodies of the body wall muscles, as previously reported (Ding and Can- dido, 2000a; Meissner et al., 2011). Y55F3BR.6 displayed expression patterns similar to those of HSP-25 and HSP-12.1. Except for the signals leaking from the neighboring body-wall muscle cells, no expression of the three sHSPs was observed in the epidermis.
