Expr4244 ser-1::gfp expression was observed in the pharyngeal muscles. In addition, ser-1::gfp expression was observed in the vulval muscles, as well as in many neurons. ser-1::gfp is not detectable in HSN or VC.
Expr12393 Pcil-7::GFP was expressed in CEM, RnB, HOB, and IL2 neurons. The translational and functional CIL-7::GFP reporter localized to cell bodies (excluding nuclei), dendrites, axons, and the cilia and ciliary bases of the extracellular vesicle (EV)-releasing sensory neurons -CEM, RnB, HOB, and IL2.
Expr15024 Consistent with the previous report on the hermaphrodites' expression, we confirmed here that for both sexes, all three of pck-1-expressing reporters express in head and body wall muscles. However, we found that the reporters also express in ventral cord and preanal neurons and nerve ring neurons per side in the head. In the tail region, both larval males and larval/adult hermaphrodites express the transgenes in the dorsal rectal ganglion and three neurons per side in the tail. The adult male shows additional expression in sex-specific neurons of the ray, postcloacal, and spicule sensilla. None of the constructs promote expression in male or hermaphrodite sex muscles, intestine, or pharyngeal muscles.
Expr14413 A GFP reporter under the control of 1,566 bp of rabs-5 upstream regulatory sequence (rabs-5p::GFP) is expressed predominantly in ciliated cells, including all dye-filling amphid and phasmid neurons, male tail ray neurons and the PQR neuron (weaker expression). No apparent differences in rabs-5p::gfp expression were found in L1 vs. adult worms.
Picture: Fig 5G, 5H, 5I.   Expr9039 The expression of nhr-206 GFP reporter transgenes starts during the comma stage of embryogenesis, and is initially seen in four unidentified cells localized in the head region. By the 2-fold stage, embryonic expression is observed in the pharynx with weaker expression in intestine. This expression pattern continues throughout all larval stages and in adults with pronounced anterior pharyngeal expression. The reporter genes were also strongly expressed in rectal gland cells, the anal sphincter, and in epidermal cells in the tail. Weaker expression was also observed in the vulva and spermatheca. In males, expression was visible in male specific neurons of the tail and rays.
Picture: Fig 5D, 5E, 5F.   Expr9037 The expression of nhr-153 GFP reporter transgenes was first detected in the 2-fold stage embryo in pharyngeal and intestinal cells. In all larval stages, the reporter gene expression was very strong in the posterior bulb of pharynx and in all intestinal cells as well as the intestinal-rectal valve or rectal gland cells. Expression was also seen in several unidentified neurons near the posterior pharyngeal bulb and in the tail. In males, very strong reporter expression was observed in some of the ray-associated neurons.
Expr11372 ADL (100%), ADF (100%), ASH (92%), RIC (88%), I2 (59%), PVR (35%), PVQ (72%), PVW (72%), HOB (25%), ray neurons (50%), protractor muscles (31%), intestine (100%), head muscle (6%). Percentages correspond to the fraction of animals examined that expressed GFP in the indicated neurons. Above 50% marked as certain, below as uncertain.
Expr9999 A 2.8 kb promoter of cysl-1 drove GFP expression mainly in the nervous system of adult hermaphrodites. The cysl-1::GFP expression pattern was similar for the transcriptional and translational reporters. GFP was observed in subsets of pharyngeal neurons, amphid sensory neurons and tail neurons, starting from late embryonic stages and persisting into adults. We identified GFP-positive cells as the AVM sensory neuron, the BDU interneurons, and the pharyngeal I1 interneurons and M2 motor neurons, based on their characteristic processes and nuclear positions. GFP in body wall muscles, hypoderm, and intestine was present in larvae but only weakly detectable in adult animals.
Expr12313 In these transgenic males, UNC-7 expression was found, in addition to the sex muscles (Expr12312), in several mating circuit neurons (SPC, PCA, PCB, HOA, and unidentified ray neurons). Similar to previous studies (Starich et al., 2009; Yeh et al., 2009; Kawano et al., 2011), this reporter also expresses in head ganglion neurons, several ventral chord neurons, and tail neurons of the hermaphrodite, in addition to few unreported pharyngeal neurons (I5, M2, I4, M3, and I1).
Expr10584 The prig-6RIG-6::GFP construct drives expression of all RIG-6 isoforms in fusion with GFP, under their endogenous promoter. rig-6 expression is detected during embryogenesis and is maintained throughout adulthood. rig-6 is expressed in I1 and I3 pharyngeal interneurons, NSM motor neurons and other unidentified head neurons. RIG-6::GFP is also localized in the cell bodies and axons of different subtypes of VNC motorneurons, in the HSN and the CAN neurons. Neuronal expression of rig-6 also includes the ALM and PLM touch receptor neurons. Non-neuronal cells expressing rig-6 include somatic muscles and the hypodermis. We also detected rig-6 expression in the developing and adult EC. The prig-6RIG-6::GFP construct drives expression of all RIG-6 isoforms in fusion with GFP, under their endogenous promoter. rig-6 expression is detected during embryogenesis and is maintained throughout adulthood. rig-6 is expressed in I1 and I3 pharyngeal interneurons, NSM motor neurons and other unidentified head neurons. We also detected rig-6 expression in the developing and adult EC.
Expr12408 With a construct including 8 kb upstream of the first exon and the entire unc-7 genomic region fused to YFP, UNC-7 expression was found, in addition to the sex muscles reported in Expr12407, in several mating circuit neurons (SPC, PCA, PCB, HOA, HOB, and unidentified ray neurons). Similar to previous studies (Starich et al., 2009; Yeh et al., 2009; Kawano et al., 2011), this reporter also expresses, in head ganglion neurons, several ventral chord neurons, and tail neurons of the hermaphrodite, in addition to few unreported pharyngeal neurons (I5, M2, I4, M3, and I1).
Reporter gene fusion type not specified.   Expr2697 In male, egl-46 expressed in FLP cells, ventral cord neurons and PVD. egl-46::cfp was expressed in the HOB neuron beginning at the L4 stage and continuing throughout adulthood. No detectable expression was seen in the HOA hook neuron.
Expr14472 The ttll-11b reporter was exclusively expressed in ciliated extracellular vesicle-releasing neurons (EVNs). Expression of GFP driven by the ttll-11b promoter was observed in the inner labial type 2 (IL2) ciliated sensory neurons in both males and hermaphrodites, as well as the male-specific PKD-2-expressing neurons (CEMs in the head; HOB and RnBs in the tail). The IL2, CEM, HOB, and RnB neurons comprise the EVNs, which release bioactive EVs to the environment.
Expr14407   In wild-type neurons, PKD-2::GFP is localized to somatodendritic (ER in the cell body and puncta along the dendrite) and ciliary regions of 4 CEM, 16 RnB, and the HOB male-specific sensory neurons. PKD-2::GFP localizes to the ciliary base (corresponding to the distal most dendrite and ciliary transition zone) in all neurons and in 56% of the cilium proper of neurons examined.
Expr14408   In wild-type neurons, PKD-2::GFP is localized to somatodendritic (ER in the cell body and puncta along the dendrite) and ciliary regions of 4 CEM, 16 RnB, and the HOB male-specific sensory neurons. PKD-2::GFP localizes to the ciliary base (corresponding to the distal most dendrite and ciliary transition zone) in all neurons and in 56% of the cilium proper of neurons examined.
Expr13485 Males containing either the fluorescent-tagged mapk-15A isoform showed expression in tail HOB and all 9 Rn CSNs. Localization of the mapk-15A isoform product was found in the cell body, dendritic projections, and ciliary regions, similar to the localization pattern observed in hermaphrodite animals.
Expr13486 Males containing either the fluorescent-tagged mark-15C isoform showed expression in tail HOB and all 9 Rn CSNs. Localization of the mapk-15C isoform product was found in the cell body, dendritic projections, and ciliary regions, similar to the localization pattern observed in hermaphrodite animals, with ciliary staining more discernable from dendritic process staining in the C-isoform.
Expr11947 A pkd-2-gfp fusion vector, that partially rescued the response phenotype of pkd-2-mutant males, was expressed in the CEM neurons, HOA, HOB, and sensory ray neurons and in the rays. Developmental regulation of expression was seen, appearing at L4 and peaking in adult males.
